چکیده

Background The Allegro XRS 20 bioreactor system is a single-use bioreactor system suitable for applications ranging from general life sciences research to seed train operations as well as small scale production at the 2 to 25 literscale. It features a 3D rectangular biocontainer that is rocked across two independent axes perpendicular to one another. These features allow for significant improvements in mixing and kLa when compared with a conventional rocking bioreactor that features a 2D “pillow” biocontainer that rocks across a single axis. The Allegro XRS 20 controller is able to control agitation of the biocontainer over a wide range of conditions, from gentle agitation required for shear sensitive cultures, to more vigorous conditions generating higher oxygen transfer rates and decreased mixing times needed for the high productivity systems seen in the biopharmaceutical industry today. Control of pH and DO is fully automated by the optical sensors supplied with the biocontainer. Gassing strategy is managed by three independent mass flow controllers and fluid additions can be made via three integrated pumps. All operations are managed through a user friendly touch screen interface.

The successful transfer of a modern CHO fed-batch process to different single-use bioreactors

Introduction Nowadays, single-use bioreactors are widely accepted in pharmaceutical industry. This is based on shorter batch to batch times, reduced cleaning effort and a significantly lower risk of cross contaminations [1,2]. One large field of the application of single-use bioreactors is the seed train cultivation of mammalian cells [1]. The focus is further extended to perform state of the a...

CFD for Characterizing Standard and Single-use Stirred Cell Culture Bioreactors

Driven by global competition and rising cost pressure in the pharmaceutical industry, over the last ten years single-use bioreactors have been increasingly used for animal cell cultivations in screening experiments, seed inoculum and seed train productions as well as in small and medium scale production processes of proteins (in particular, antibodies and vaccines). In contrast to re-usable bio...

High cell density cultivation and recombinant protein production with Escherichia coli in a rocking-motion-type bioreactor

BACKGROUND Single-use rocking-motion-type bag bioreactors provide advantages compared to standard stirred tank bioreactors by decreased contamination risks, reduction of cleaning and sterilization time, lower investment costs, and simple and cheaper validation. Currently, they are widely used for cell cultures although their use for small and medium scale production of recombinant proteins with...
